Mouse brain-type cannabinoid receptor (CB1) cDNA was cloned and sequenced. Comparison to rat and human CB1 sequences showed extensive homology (>96%) both at the nucleotide and protein levels implicating the importance of this protein in mammalian brain physiology.
To develop a simple, standardised procedure for antifungal susceptibility of yeasts, disc diffusion test was evaluated in comparison to agar dilution and the macrobroth dilution procedure of the National Committee for Clinical Laboratory Standards USA. One hundred Candida strains isolated from clinical material were used against four antifungal agents (amphotericin B, 5-flucytosine, ketoconazole and fluconazole). Inoculum size of 10(6) yeasts/ml just failed to produce confluent colonies at 37 degrees C after 48 h on yeast nitrogen-glucose-agar with or without asparagine medium. The zones of inhibition were correlated with both broth and agar dilution minimum inhibitory concentrations (MICs). The results were found to correlate well using amphotericin B, 5-flucytosine and fluconazole (with broth dilution only). But there was no significant correlation using ketoconazole. Therefore, disc diffusion susceptibility testing of Candida strains appears to be generally applicable except for azoles where broth dilution test would confirm the resistant isolates.
A pharmacodynamic interactional study with omeprazole was undertaken in rats. Omeprazole (7 mg/kg, orally once daily for 14 days) significantly prolonged pentobarbitone (30 mg/kg, ip) induced hypnosis while it had no effect on haloperidol (1 mg/kg, ip) induced catalepsy or morphine (5 mg/kg, ip) induced analgesia models in rats. The study highlighted the fact that dynamic interaction with omeprazole was selective.
Chemoautotrophic nocardioform (CAN) bacteria had been repeatedly isolated from fish with ulcerative disease syndromes (EUS) from the massive epizootics that had repeatedly occurred since 1988 in eastern India as the major or only pathogenic agent in the background of distinctive environmental and epizootic data. Since these isolates bear significant similarity to the human and rat leprosy bacilli, attempts had been made to demonstrate the pathogenicity of this fish pathogen in the "Swiss" strain of mice as a convenient model. The studies reveal that the fish CAN bacteria could produce pathogenic effects in mice similar to that of the rat leprosy bacillus.

Nosocomial disseminated candidiasis was diagnosed in 6 out of 200 (3%) children receiving pediatric intensive care over a period of 9 months. The ages of patients ranged between 20 days to 3 years; 4 were < 2 months. Therapy with broad spectrum antibiotics (in all), indwelling cannula (in all), peritoneal dialysis (in 3), low birth weight (in 3) and invasive hemodynamic monitoring were recognizable predisposing factors. The diagnosis was suspected on an average after 14 days, PICU stay (range 8-20 days). All the patients showed a secondary worsening after evidence of improvement from the primary illness. It was characterized by lethargy, fever (in 3), weight loss (in 3), loose stools (in 2) and respiratory distress (in 3), and was indistinguishable from any bacterial sepsis. Presumptive diagnosis was made on basis of KOH wet mount and Gram stained smear findings of mycelia, and was confirmed later on isolation of candida species from one or more body sites and blood culture. All the patients showed disappearance of symptoms and mycological cure within 6-14 days of oral itraconazole therapy, (10 mg/ kg/day in 2 divided doses). The therapy was continued for upto 14 days after sterile fungal blood culture, and was well tolerated. Fungal superinfection especially with candida must be looked for in hospitalized patients suspected of nosocomial infection. Early oral itraconazole is effective in disseminated candidiasis and well tolerated by children.

Immunoblot detection of antibody against 47 KD cytoplasmic antigen of Candida albicans was evaluated in diagnosis of invasive candidiasis and compared to whole cell agglutination and gel diffusion tests for detection of anticandidal antibody in 64 patients. The patients included 17 with culture proved candidemia, 34 with significant candiduria (more than 10,000 colony forming units per ml of urine) and 13 with nonsignificant candiduria. Antibody against 47 KD antigen was found to be the best indicator for diagnosis of invasive candidiasis even in patients with malignancy. The sensitivity of this procedure was 82.4%, specificity 86.7%, positive predictive value 77.8%, negative predictive value 89.7% and efficacy 85.1%. The gel diffusion procedure lacked in sensitivity whereas whole cell agglutination lacked in specificity. Detection of antibody against 47 KD antigen proved to be a valuable adjunct in the diagnosis of invasive candidiasis.
