An evaluation of primary enamel pretreatments and their effects on resin retention.

The effects of various enamel pretreatments on bond strengths between primary enamel and resin were investigated. Pretreatments included: removal of the outer enamel surface, varied exposure to phosphoric acid, and the use of an organic coupling agent. Results indicated highest bond strengths utilizing acid pretreatments and an organic coupling agent.

[Segmental osteotomies of the mandible in the orthodontic surgical treatment of malformations of the facial bones].

In this article the Authors take under consideration surgical techniques of segmental osteotomy of the mandible in the treatment of the maxillo-facial skeleton alterations. The anterior inferior alveolar segmental osteotomy, total subapical osteotomy, anterior subapical osteotomy, genioplasty and the midline mandibular osteotomy are presented with their surgical techniques and indications. Therefore the authors present two cases of patients undergone to orthodontic--surgical treatment.

Immunochemical determination of lonidamine in rat tissues and blood serum.

Lonidamine (LND) is an antitumor drug which interferes selectively with the energy metabolism of neoplastic cell. Because of its physico-chemical properties, LND determination with conventional methods, i.e. high performance liquid chromatography and spectrofluorimetry, gives rise to several problems: LND is a lipotropic drug which becomes intimately associated with biological membranes so that it is impossible to extract all the drug bound, thus leading to an underestimation of the LND content in cells and tissues. These difficulties can be overcome by the immunoenzymatic method described here. The assay is simple, rapid, practical, highly sensitive (2-5 ng/ml) and particularly suitable for the analysis of multiple samples (twelve samples in triplicate for each plate). There is, moreover, a great improvement in data reproducibility.

Lonidamine-induced outer membrane permeability and susceptibility of mitochondria to inhibition by adriamycin.

Low concentrations of Adriamycin (ADM) do not inhibit the oxygen consumption of rat liver mitochondria because of the inability to cross the outer membrane. The involvement of this membrane as a permeability barrier is demonstrated by the results with exogenous ferrocytochrome c. ADM does not affect the basal rate of ferrocytochrome c oxidation, which, on the contrary, increases in Lonidamine (LND)-treated mitochondria. This difference lies in the capacity of LND to unmask the redox carriers in the inner membrane, i.e. cytochrome c:oxygen oxidoreductase which is also a marker of the outer membrane permeabilization. Therefore, the enhancement of ADM's effect on mitochondrial respiration by LND must be ascribed to its permeabilizing effect on the outer mitochondrial membrane.

Late-onset 3 beta-ol-hydroxysteroid-dehydrogenase deficiency: prevalence, clinical presentation and endocrine pattern.

3 patients with late-onset 3 beta-HSD deficiency are described. They presented increased DHEAs/delta 4 ratio either in basal conditions or after ACTH stimulation. All patients were hirsute but only one had increased serum values of delta 4 and total testosterone. However, all patients presented increased serum levels of free testosterone. Two patients had normal menstrual cycles and in one case a secondary PCO was found.

Enhancing effect of lonidamine on the inhibition of mitochondrial respiration by adriamycin.

Pretreatment of rat liver mitochondria with lonidamine (LND), 1-(2,4 dichlorobenzyl)-1 H-indazol-3-carboxylic acid, increases their susceptibility to respiratory inhibition by adriamycin (ADM). Because LND does not inhibit the rate of oxygen consumption at the concentrations used, its potentiating effect may be exerted by favoring the interaction of ADM molecules with their sites in the inner mitochondrial membrane. LND does not modify the permeability on the inner membrane for large molecules. However, its enhancing effect can be ascribed to a modified permeability of the outer mitochondrial membrane which may represent a barrier for ADM entry.

In vitro and in vivo potentiation by lonidamine of the antitumor effect of adriamycin.

Lonidamine (LND), a powerful antispermatogenic compound, has been shown to impair differentially the energy metabolism of normal and neoplastic cells and to increase significantly the survival of animal bearing experimental tumors. Clinical trials are presently ongoing and preliminary favourable results have been obtained with LND as a single agent or associated with other antineoplastic drugs. In this work the effect of the combination LND plus ADM was investigated on in vitro and in vivo tumors. The in vitro effect was analysed by the colony forming technique and the in vivo antitumor activity was assessed by means of local control parameters, increase in host lifespan and weight reduction of metastasing target organs (liver, kidney, ovary). The results obtained indicate that LND is poorly effective when used alone, while a very high cytotoxicity is observed when it is combined with ADM. Exposure of cells to different time-sequences and schedules of both drugs elicits a different response. In particular a synergistic effect is obtained when ADM precedes LND. These results have been preliminarily confirmed by in vivo studies.
