Effect of platelets on apparent leucocyte ascorbic acid content.

The leucocyte ascorbic acid content is widely used as a measure of tissue ascorbic acid status. Standard methods of analysis, however, isolate both leucocytes and platelets (buffy layer), with consequent overestimation, since platelet ascorbic acid is attributed to the leucocytes. Fourteen healthy individuals on ascorbic acid supplements and 11 patients on mega dose ascorbic acid therapy were studied. A significant correlation was demonstrated between the 'leucocyte' ascorbic acid content and the platelet: leucocyte ratio (r = 0.70, P < 0.001). It is suggested that changes in the relative distribution of platelets and leucocytes in the blood will result in an apparent change in the 'leucocyte' ascorbic acid content regardless of any actual change in the ascorbic acid content of the cells.

Nomenclature of transposable elements in prokaryotes.

Transposable elements are defined as specific DNA segments that can repeatedly insert into a few or many sites in a genome. They are classified as simple IS elements, more complex Tn transposons and self-replicating episomes. Definitions and nomenclature rules for these three classes of prokaryotic transposable elements are specified.

Acute reactions to mega ascorbic acid therapy in malignant disease.

Three cases are described, 2 of Hodgkin's disease and a further case of bronchial carcinoma, where high dosage ascorbic acid treatment appeared to be associated with the development of potentially dangerous symptoms. It is suggested that mega ascorbic acid therapy should be given with caution in malignant disease, with a slow build-up over several days to high levels of dosage.

Development, survival, and oviposition of the rabbit tick, Haemaphysalis leporispalustris (Packard) (Acari: Ixodidae), at constant temperatures.

The relationship between development, survival, and oviposition rates, and five constant temperatures is described for the developing and reproductive life stages of the rabbit tick, Haemaphysalis leporispalustris (Packard). Development was most rapid at 29.7 C for egg and larvae, and 35.6 C for nymphs. Survival was greatest between 14.7 and 29.7 C, but was reduced at 35.6 C for eggs and larvae. Oviposition was optimal between 19.9 and 29.7 C with a maximum mean daily oviposition of 253 eggs/female/day on the third day of oviposition at 29.7 C. Maximum mean total eggs per female produced was 1,157 with a range of 277 to 3,327. The total number of eggs produced per female correlated with the weight of the engorged female. Both linear and sigmoid curve equations were used to approximate the relationship between the temperature and development rates of eggs, larvae, and nymphs. The nymphs had the highest heat requirements of the 3 development stages. Comparisons with field host-parasite data indicate that the rabbit tick has a 2-yr life cycle in southwestern Nova Scotia.

Specificity of DNA-associated nuclear antigens in HeLa cells and distribution during the cell cycle.

Chromosomal nonhistone protein:DNA complexes prepared from synchronized HeLa cells were used to immunize white rabbits. The antisera reacted specifically in complement fixation tests with chromatins isolated from HeLa cells and not with those from a number of other human and animal tissues. Specificity to this cell type was also demonstrated by immunocytochemical reaction. Both immunochemical tests revealed that the specific antigens are continuously present throughout the cell cycle. The immunological activity was dependent upon chromosomal nonhistone protein(s) being bound to DNA. Our findings are consistent with these chromatin antigens being stable nuclear components [complexes of chromosomal non-histone protein(s) with DNA] characteristic of cellular differentiation.

Inhibition of 5,7-dihydroxytryptamine-induced supersensitivity to 5-hydroxytryptophan in mice by treatment with cycloheximide.

Intracisternal injection of 5,7-dihydroxytryptamine (5,7-DHT) following treatment with desmethylimipramine induced development of behavioral supersensitivity to the intraperitoneally administered serotonin precursor 5-hydroxytryptophan (5-HTP) in the mouse. This behavioral syndrome, characterized by tremor and muscle twitches (myoclonus), showed a clear dose-response relationship with 5,7-DHT as well as with 5-HTP. Mice lesioned with a low dose of 5,7-DHT (20 micrograms) or a placebo were treated repeatedly with a protein synthesis inhibitor, sycloheximide (45 mg/kg, s.c., every 12 h for up to 10 days). This treatment resulted in a reversible decrease of cerebral protein synthesis varying between 70 and 20% with time between treatments. The myoclonic response to 5-HTP in animals pretreated with 5,7-DHT and by cycloheximide showed a decrease in intensity within 24 h when evaluated quantitatively by an electronic activity monitor, the results of which were confirmed by direct observation. Cycloheximide also exerted a similar, though smaller, effect following full development of sensitivity to 5-HTP over 10 days. These effects may de mediated by inhibition of rapidly turning over serotonin receptor proteins, although their interpretation is somewhat obscured by possible toxic effects of cycloheximide.

Deficits in visual learning produced by posterior temporal lesions in cats.

Eight cats with lesions in the posterior temporal (PT) cortex, seven cats with lesions in the basolateral amygdala (BLA), and eight intact controls were observed on eight tests of visual discrimination learning and of spontaneous responses to salient visual stimuli. The effects of the two lesions were somewhat dissociable. The PT lesions were accompanied by a severe deficit in pattern discrimination learning but no loss in visual tracking or orientation to the silhouette of a threatening cat. The BLA lesions produced a milder and less consistent loss in pattern discrimination but serious defects in tracking and reponse to the cat silhouette. Both operated groups performed well on the visual cliff. The deficit from PT lesions appeared independent of damage to the geniculocortical system. The parallel of symptoms from PT lesions in cats and inferotemporal lesions in monkeys is discussed.

Bodily communication and personality.

Fifteen measures of non-verbal communication were coded from videotaped interactions between a female confederate and 18-21 year old female subjects (n = 46). Three measures of extraversion and neuroticism had previously been taken from the subjects, as was a measure of IQ. Correlation and factor analysis revealed significant relationships between certain of the variables. Extraversion was strongly associated with speaking more; a teacher's rating of neuroticism was associated with touching the self, pausing during conversation, and an absence of expressive gesture; lower IQ was associated with smiling while listening; and self-report personality questionnaire neuroticism was associated with gaze aversion.

Repression of biotin biosynthesis in Escherichia coli during growth on biotin vitamers.

A strain of Escherichia coli in which the lacZ gene was fused to the bioA promoter was constructed. Colonies of this strain formed Lac(+) colonies on low-biotin agar (1.6 to 4.1 nM) and Lac(-) colonies on high-biotin agar (41 nM). This lac-bio fusion strain was used to study the question of whether cells growing on the biotin vitamers d-biotin-d-sulfoxide (BDS) and dethiobiotin (DTB) generate enough biotin to give maximal repression of beta-galactosidase synthesis. Repression by high concentrations (400 nM) of BDS was almost maximal (about 96%), whereas DTB repression reached a saturation level of about 80% with increasing DTB concentrations. The levels of repression obtained with both vitamers were sufficient to cause the colonies to appear Lac(-). When the lac-bio fusion was transduced into lines carrying mutations (bis) that prevent reduction of BDS to biotin, the transductants were not repressed by added BDS. Repression by BDS is unlikely to result from accumulation of extracellular biotin-related substances because (i) washed bis(+) cells were not detectably derepressed when transferred into medium containing BDS and (ii) washed bis cells were not detectably repressed when transferred into medium in which bis(+) cells had grown. Lactose agar plates containing high concentrations of DTB or BDS comprise an efficient selective medium for bioB or bis mutants and were used to isolate spontaneous mutations of these genes. This method should be adaptable to the selection of mutations in any biosynthetic pathway subject to end-product repression.
