Changes in plasmatic biochemical parameters during experimental murine paracoccidioidomycosis.

Determinations of plasma cholesterol, HDL-cholesterol, triglycerides, total proteins, bilirubin, aspartate transaminases (AST), glucose, iron and total iron binding capacity were evaluated after 15 and 90 days of intraperitoneal infection with 5 x 10(6) virulent yeasts of Paracoccidioides brasiliensis Pb 18 isolate in resistant (A/J) and susceptible (B10.A) mice. Significant differences between control and infected groups were seen: an increase of triglycerides and total proteins for both strains, a decrease of glucose and HDL-cholesterol in B10.A mice and of iron in both strains, while AST, bilirubin and TIBC increased only in B10.A mice. While glucose, AST, bilirubin and TIBC were strongly modified in susceptible mice, no differences were seen in the resistant infected group, thus confirming the usefulness of biochemical parameters as an index of infection severity, as well as of recovery.

Direct detection of Bordetella pertussis and Bordetella parapertussis: comparison of polymerase chain reaction and culture.

We evaluated the diagnostic performance of a genomic DNA amplification method for Bordetella pertussis and Bordetella parapertussis compared with culture isolation. Aliquots from B. pertussis and B. parapertussis cultures were added to sterile physiological saline or sterile distilled water to give bacterial suspensions of 10(8) cells/ml and serial dilutions were prepared. Suspensions in physiological saline were cultured on charcoal agar medium; bacterial growth was observed up to dilutions of 10(-7). Suspensions in distilled water were subjected to DNA extraction and nested polymerase chain reaction (PCR) was performed on the extracts; the PCR was positive up to dilutions of 10(-8) for B. pertussis and 10(-9) for B. parapertussis. Since the efficacy of culture isolation, regarded as the standard for the detection of B. pertussis and B. parapertussis, declines after the first stage of pertussis or with prior vaccination or antibiotic therapy, PCR, although not yet standardized, may provide an alternative diagnostic tool.

[Generalized lymphatic dysplasia in a 6-month-old nursing infant].

G.L.D. is a rare syndrome characterized by chylothorax, chylous ascites and lymphedema associated to minor symptoms following from lymphangiectasis or to lymphangiomatosis. This syndrome is caused by congenital dysplasia of lymphatic vessels and has, generally, an extremely severe prognosis. In the present papers the authors describe one case of G.L.D. in a six months old infant with chylous ascites, chylothorax, lymphedema, hemangioma and a history of fetal ascites. The preservative (dietetic and evacuative) therapy was not resolutive, therefore an exploratory laparotomy was necessary, but no surgical solution was possible because of the absence of chylous vessels in the anatomical region of the principal lymphatic route. However, laparatomy led to a progressive recovery of ascites and chylothorax, probably due to thrombosis of dysplasic lymph vessels and to neoformation of collateral circulation.
