[Contribution to the study of labeled neurohypophysial hormones. Preliminary data on the properties of the macromolecular compound: vasopressin-dextran (author's transl)].

The neuropeptide binding sites at the level of their target cells can be localized by several morphological methods. For one of the neurohypophysial hormones, vasopressin, not all labelling techniques can be applied: radioiodinated vasopressin has no biological activity; only 3Hvasopressin can be used. Immunocytochemical evidence for vasopressin receptors must be supported by complementary data in order to demonstrate the specificity of the binding. A new labelled ligand is prepared: vasopressin-dextran. In vivo, this macromolecular compound has antidiuretic properties equivalent to those of free vasopressin. In vitro, it gives a cross reaction with specific anti-vasopressin antibodies. For the same macromolecular compound, the ratio between the conjugated vasopressin estimated through R.I.A. and that assayed by biological method was 30.

[Postnatal changes in the content of neurohypophyseal enzymes in the Brattleboro rat].

The development of the neurohypophysial content of vasopressin and oxytocin in the Brattleboro rats is investigated. During the early development stages, no alteration of the oxytocin content could be observed owing to the reduced synthesis of vasopressin in the heterozygous rats (Di/+) or to the lack of this hormone in the homozygous rats (Di/Di).

Infundibular localization of vasopressin, oxytocin and neurophysins in the rat; its relationships with corticotrope function.

The distribution of vasopressin (VP), oxytocin (OXY) and neurophysins 1 and 2 (N1, N2) has been studied in the median eminence (ME) of normal, heterozygous (HDI) and Brattleboro (DI) rats. Numerous thin periportal VP and N2 fibres exist in the normal and HD1 rats; they have never been observed in the DI rats. N1 and OXY fibres in the external layer of the median eminence are rare. Adrenalectomy increases periportal VP and N2 loading in normal and HDI rats; it does not modify the appearance of the DI median eminence. Dexamethasone prevents external VP and N2 overloading in adrenalectomized rats injected during the whole postoperative period. Injections of vasopressin indicated that it had a negative feedback effect during a short time (3 days) but not during a longer period (7 days). The suprachiasmatic neurons are stained only by anti-VP and anti-N2 antibodies. Their overstaining induced by adrenalectomy disappears with dexamethasone, aldosterone or vasopressin treatment. This central effect of hormones is not necessarily associated with disappearance of overloading in the external layer of the ME. The hypothalamo-infundibular tract carrying VP and N2 is involved in regulatory mechanisms of the corticotrope axis. Its relationships with suprachiasmatic neurons are discussed.

Evidence of vasopressin in adenohypophysis: research into its role in corticotrope activity.

The presence of vasopressin (VP) in pars distalis of rats and pigs was investigated. Using radioimmunoassay and bioassay of VP, a substance with immunological and biological properties of this hormone was found. This substance was not detected in the adenohypophysis of rats with diabetes insipidus. A partial purification of the VP-like peptide showed that it had the chromatographic and electrophoretic properties of VP. It could be identified with arginine vasopressin (AVP) in the case of the rat and lysine-vasopressin (LVP) in the case of the pig. In the Wistar strain, adrenalectomy induced progressively increasing concentrations of adenohypophysial VP. This increase was significant 15 days after surgery. It could be prevented by treatment with dexamethasone. These results indicate that the presence of VP in the anterior pituitary is related to the regulation of ACTH secretion.

[Immunoreactive vasopressin in the supra-chiasmatic nucleus. Preliminary data in rats].

The immunohistochemical technique for the vasopressin localisation shows the presence of this hormone in neurons of the rat supra-chiasmatic nucleus. Different experimental conditions induce modifications of the supra-chiasmatic vasopressin, and, in the same time, the storage of immunoreactive peptide in the fibres terminating in the external zone of the median eminence. However, the supra-chiasmatic, supraoptic and paraventricular changes are not obligatory similar.

[Changes in rat hypothalamo-neurohypophyseal system after repeated ethanol administration].

The histochemical, immunohistological and histoenzymatic study of the hypothalamo-neurohypophysial system of the rat shows that chronic ethanol administration induces: a temporary blockage of vasopressin synthesis, a vasoconstriction of the neurohypophysial capillaries, a dendritic storage of immunoreactive vasopressin. In our experimental conditions, a long ethanol treatment disturbs the balance between vasopressin synthesis and release.
