Binding of heparin to human microvascular endothelial cells and the effect on proliferation.

We have studied the binding of 125I-heparin to human omental microvascular endothelial cells (HOME cells) and investigated its effect on cell proliferation. At 20 degrees C, the binding reached a steady state from 4 hours onwards. Saturation of 125I-heparin binding occurred at 200 nM. Scatchard analysis indicated one class of binding sites (KD = 0.023 microM, 2 X 10(-6) sites/cell). Using fractionated heparin and other sulfated polysaccharides it was demonstrated that the binding was dependent on the charge and the molecular weight of the compounds. The binding was followed by the partial internalization of the bound ligand (23.8%). Heparin and Stipocus Japonicus mucopolysaccharide (SJAMP) inhibited the proliferation of exponentially growing HOME cells and human umbilical vein endothelial cells (HUVE cells). However, serum-deprived HOME cells were not inhibited by heparin.

Natural proteinase inhibitors: blood coagulation inhibition and evolutionary relationships.

1. Natural proteinase inhibitors are divided into polysaccharides, plasma proteinase inhibitors and natural non-plasma inhibitors. 2. Polysaccharides are antithrombin-III and heparin co-factor-II dependent or independent regarding their biological activity. Knowledge of the inhibitory mechanism at a molecular level was gained by the study of heparin. 3. Antithrombin-III, heparin-co-factor-II and alpha 2-macroglobulin are the most important plasma proteinase inhibitors involved in coagulation. alpha 2-macroglobulin has a particular inhibitory mechanism. 4. Non-plasma proteinase inhibitors were isolated from many species. They inhibit mainly the contact activation and fibrinolysis. 5. The evolutionary relationships are poorly understood.

Cefotaxime in human lung tissue.

In the course of thoracic surgery in 79 patients lung tissue samples were taken up to around 2 and 4 h, respectively, after i.v. injection of 1 (n = 22) or 2 g cefotaxime (n = 57). In these samples cefotaxime levels were measured using a microbiological method. About 1 h after injection 1.6 and 5.1 microgram/g (median values), respectively, were found. Even later after injection cefotaxime could still be detected. These results are evaluated with respect to the problem encountered in the biological measurement of concentrations of ester cephalosporins in tissue due to desacetylation by hemolytic samples.

[African histoplasmosis of the lung (author's transl)].

Lung resection showing giant cell (African) histoplasmosis was performed because of suspected lung tumour in a patient who had lived mainly in West Africa. A primary focus or signs of localisation in other organs were suspected radiographically in the maxilla and in the skeleton. However, they could not be confirmed as the patient declined further investigations for diagnosis and therapy.
