Polyamines in Rice Seedlings under Oxygen-Deficit Stress.

Incubation of 3-d-old seedlings of Oryza sativa L. cv Arborio under anaerobic conditions, leads to a large increase in the titer of free putrescine while aerobic incubation causes a slight decrease. After 2 days, the putrescine level is about 2.5 times greater without oxygen than in air. The rice coleoptile also accumulates a large amount of bound putrescine and, to a lesser extent, spermidine and spermine (mainly as acid-soluble conjugates). Accumulation of conjugates in the roots is severely inhibited by the anaerobic treatment. Feeding experiments with labeled amino acids showed that anoxia stimulates the release of (14)CO(2) from tissues fed with [(14)C]arginine and that arginine is the precursor in putrescine biosynthesis. After 2 d of anoxia, the activity of arginine decarboxylase was 42% and 89% greater in coleoptile and root, respectively, than in the aerobic condition. The causes of the differences in polyamine metabolism in anoxic coleoptiles and roots are discussed.

Expression of c-jun protooncogene in human myelomonocytic cells.

A prototypic "immediate early" gene, c-fos, has been extensively investigated in relation to the differentiation and activation of myelomonocytic cells. The c-fos gene product is associated in transcriptional complexes with the c-jun product. These protooncogenes are part of the regulatory network of gene expression. The present study was designed to investigate expression of the c-jun protooncogene in human circulating myelomonocytic cells. We found that c-jun is constitutively expressed in normal monocytes and granulocytes, whereas low levels of transcripts are found in lymphocytes. Acute myelogenous leukemia (AML) samples of French-American-British Cooperative Group (FAB) subtypes 1 through 4 express appreciable levels of this protooncogene. Normal phytohemagglutinin (PHA)-activated lymphocytes express high levels of c-jun. Expression in normal myelomonocytic cells is detectable even after 18 hours of culture. The c-jun transcripts in myelomonocytic cells have a half-life of approximately 20 minutes and are superinduced by cycloheximide, which affects both the degradation rate of mRNA and the transcriptional activity of the c-jun gene. Functional activation of monocytes and granulocytes with phorbol esters, lipopolysaccharide, and tumor necrosis factor (TNF) increase c-jun expression. This induction is rapid, transient, and does not require intervening protein synthesis. Runoff experiments showed that in freshly isolated untreated monocytes, the c-jun gene is constitutively transcribed, and that induction by lipopolysaccharide is at least in part at the transcriptional level. Moreover, lipopolysaccharide (LPS) treatment reduced the degradation rate of c-jun transcripts, prolonging the half-life to approximately two hours. Expression of c-jun in resting and activated monocytes and granulocytes suggests that this protooncogene may play a role in the differentiation and activation of cells belonging to the myelomonocytic lineage.

Clinical assessment of sulbenicillin in acute respiratory tract infection.

The effectiveness of sulbenicillin was assessed in the treatment of 15 adults, 12 suffering from bronchopneumonia and three from lobar pneumonia. Six patients had concomitant complications. At a parenteral dose of 2 g twice or thrice daily for at least seven days, all patients became afebrile by the sixth day, and chest X-rays became normal in nine patients and improved in five. It is concluded that sulbenicillin, which was well tolerated, is clinically effective in the treatment of severe respiratory tract infections, also in patients with major impairment of their immune response systems.

[Tibial periostal graft in the repair of the primary cleft palate: preliminary report of a new technique (author's transl)].

Reconstruction of the oral plane of a primary cleft palate using tibial periostal graft at the primary repair stage seems a highly appealing technique: -- the osteogenic power of the periosteum used as a free transplant has been demonstrated long ago (Ollier, Axhausen); -- two requisites must be filled when primary palate repair is undertaken: maintainance of the nasal fossa size and maintenance of the palatal fibromucosa that must be not submitted to any traction whatsoever, otherwise endognathia will arise. A vast, tight and strong periostal plane will be obtained with the periostal graft, the latter bringing also bone to the cleft. Better still, such a periostal graft can be extended backwards over the entire length of the cleft bony palate which will subsequently create a situation requiring simple soft palate repair.

Experiences with 197HgCl2 in the diagnosis of head and neck malignancies.

197 Hg Cl2 (mercuric dichloride) was used in the diagnosis of 30 cases of epidermoid carcinoma of the head and the neck, and in 5 cases of benign tumours and non-neoplastic lesions. The results show that radioisotope scanning is not a certain method of diagnosis but that it affords supplementary information essential to correct therapy.
