An enzyme-linked immunosorbent assay, using an EDTA-extracted antigen for the serology of Haemophilus pleuropneumoniae.

An enzyme-linked immunosorbent assay (ELISA) was proposed as an alternative to the complement-fixation test (CF) for the detection of antibodies of Haemophilus pleuropneumoniae, agent of the pleuropneumonia in pigs. In tests done with different antigen-extraction procedures (including Tween 20, sodium dodecyl sulfate, aqueous phenol, sonification, and heat treatment at 120 C), ethylenediaminetetraacetic acid (EDTA) provided a satisfactorily reactive antigen. Chromatography purification on Sephacryl S200 improved the specificity of this antigen. Using hyperimmune rabbit sera, we investigated the specificity and the sensitivity of the ELISA with the EDTA-purified antigen of the different serotypes of H pleuropneumoniae on selected swine sera in herds with confirmed H pleuropneumoniae infection, from specific-pathogen-free animals showing doubtful CF reactions. The ELISA proved to be highly specific and more sensitive than the CF test. Furthermore, evidence of cross-reactions with H parasuis, a common bacteria isolated in swine populations, was not found.

[Maturation curve of the ultrasonographic alpha angle according to Graf's untreated hip joint in the first year of life].

In addition to Graf's classification into different hip-types a maturation-curve of the sonographic alpha-angle was established in order to optimize the differentiation between mature hip joints and those ones which need follow-up and those ones which need treatment at any time within the first year of life. The results obtained are confirming our present knowledge about the spontaneous maturation of physiologically immature hip joints: The mean-value of the alpha-angle crosses the 60 degrees-borderline at about the age of two months, then reaches an about 64 degrees-level at about four months. This 64-degrees-level is more or less unchanged up to the end of the first year of life.

Polarization-sensitive optical coherence tomography of dental structures.

Optical coherence tomography (OCT) has been developed during the last 10 years as a new noninvasive imaging tool and has been applied to diagnose different ocular and skin diseases. This technique has been modified for cross-sectional imaging of dental structures. In this first preliminary study the technique was applied to obtain tomographic images of extracted sound and decayed human teeth in order to evaluate its possible diagnostic potential for dental applications. Classical OCT images based on reflectivity measurements and phase retardation images using polarization-sensitive OCT were recorded. It was demonstrated that polarization-sensitive OCT can provide additional information which is probably related to the mineralization status and/or the scattering properties of the dental material. One of the attractive features of OCT is that it uses near-infrared light instead of ionizing radiation. Furthermore, high transversal and depth resolution on the order of 10 microm can be obtained. Present limitations, e.g. the limited penetration depth, and possible solutions are discussed.

[Computer-assisted detection of cerebral embolism using a multi-range principle].

Since its introduction into the clinical setting at the beginning of the 'eighties, transcranial Doppler sonography (TCD) has gained increasing acceptance as a non-invasive neuromonitoring technique. The ability of TCD to detect embolic particles within the cerebral blood vessels has been the subject of intensive investigation in recent years. In the present paper, a new method for the detection of emboli (multirange principle), intra-operatively during a neurosurgical intervention in a 35-year-old patient with a tumour in the posterior fossa and an open foramen ovale, is described for the first time. The method opens up new possibilities for the intra-operative management and the rapid assessment and diagnosis of paradoxical cerebral embolism.
