Isolation and characterization of the cDNA clone and genomic clones of a new HLA class II antigen heavy chain, DO alpha.

From a human cDNA library constructed from a consanguineous HLA-homozygous cell line, AKIBA (HLA-A24, Bw52, DR2, Dw12, DQw1, and Cp63) (Cp63, a new SB type), a cDNA clone encoding a new HLA class II antigen heavy chain named DQ alpha was isolated, and was analyzed by Southern blot hybridization and by nucleotide sequence determination. The nucleotide sequence of the DO alpha cDNA was distinct from those of the DR alpha, the DQ alpha, and the DP alpha cDNA, but showed some characteristic features of the class II antigen alpha-chains. We also isolated and identified genomic clones specifying the DO alpha gene. Genomic analyses of cell lines with different HLA-DR serotypes with the use of the DO alpha cDNA as a probe indicated the existence of a single DO alpha gene that exhibited little restriction enzyme polymorphism.

The mechanism of decreased Na+-dependent D-glucose transport in brush-border membrane vesicles from rabbit kidneys with experimental Fanconi syndrome.

In our previous paper (Yanase, M. et al. (1983) Biochim. Biophys. Acta 733, 95-101) we reported that the Na+-dependent D-glucose uptake into brush-border membrane vesicles is decreased in rabbits with experimental Fanconi syndrome (induced by anhydro-4-epitetracycline). In the present paper we investigate the mechanism underlying this decrease. D-Glucose is taken up into the osmotically active space in anhydro-4-epitetracycline-treated brush-border membrane vesicles and exhibits the same distribution volume and the same degree of nonspecific binding and trapping as in control brush-border membrane vesicles. The passive permeability properties of control and anhydro-4-epitetracycline-treated brush-border membrane vesicles are shown to be the same as measured by the time-dependence of L-glucose efflux from brush-border membrane vesicles. D-Glucose flux was measured by the equilibrium exchange procedure at constant external and internal Na+ concentrations and zero potential. Kinetic analyses of Na+-dependent D-glucose flux indicate that Vmax in anhydro-4-epitetracycline-treated brush-border membrane vesicles (79.3 +/- 7.6 nmol/min per mg protein) is significantly smaller than in control brush-border membrane vesicles (141.3 +/- 9.9 nmol/min per mg protein), while the Km values in the two cases are not different from each other (22.3 +/- 0.9 and 27.4 +/- 1.8 mM, respectively). These results suggest that Na+-dependent D-glucose carriers per se are affected by anhydro-4-epitetracycline, and that this disorder is an important underlying mechanism in the decreased Na+-dependent D-glucose uptake into anhydro-4-epitetracycline-treated brush-border membrane vesicles.

67Ga-binding substances in abscess and normal tissues.

Abscess-induced animals and normal animals were treated with 67Ga-citrate. Abscess, kidney, heart, lung, and spleen were excised and homogenized. After removal of the nuclear fraction, each of these homogenates was digested with protease. After digestion, the supernatants of the reaction mixtures were applied to a Sephadex G-100 column. Resultant eluates were analyzed for radioactivity, protein, uronic acids, and sialic acids. Sodium sulfate-35S was administered to animals that were then treated by the same procedure as that followed for animals treated with 67Ga-citrate. In abscess, kidney, lung, heart, and spleen, sizeable amounts of 67Ga had been bound to the sulfated acid mucopolysaccharides with molecular weights of about 10,000, and to the sulfated acid mucopolysaccharides, a species whose molecular weights exceeded 40,000. Based on the results presented here, it is clear that 67Ga-binding substances in abscess and also in the above four organs are sulfated acid mucopolysaccharides.

Radioactive implant induced x-ray emission technique for noninvasive determination of iodine content in thyroid: experimental study.

A new technique, radioactive implant x-ray emission spectrometry to determine the in vivo iodine content of the human thyroid is proposed. The variations of counting rate and effective excitation efficiencies of I K alpha (28.6 keV) with iodine content, thyroid volume and skin-thyroid distance were studied for the 201Tl source to examine the properties of excitation and photon attenuation. As a result, the gland depth and volume could be estimated from the peak ratios of 30.7 keV/167.6 keV and 28.6 keV/167.6 keV. Using a 1 MBq 201Tl implant source, the detectable minimum iodine concentration found was approximately 70 micrograms/g for 2000 s measuring time in the phantom experiment. The effectiveness of the RIXE technique is discussed. It was concluded that the 201Tl RIXE spectrometry might serve to determine the in vivo iodine content of the human thyroid.

[Two cases of primary intracranial malignant lymphoma].

Two cases of primary intracranial malignant lymphoma are reported. Case 1 was a 65-year-old female who was operated on for a left frontoparietal tumor in 1966 at our hospital, and the tumor was diagnosed as reticulum cell sarcoma histologically. Irradiation was also done. Thereafter, she had had no serious trouble except for slight right hemiparesis. In March of 1982, right hemiparesis became worse and disturbance of consciousness also appeared. CT scan and left carotid angiogram revealed a left fronto-temporoparietal tumor. Subtotal removal of the tumor was performed on the 1st of April. The tumor was diagnosed as malignant lymphoma, of large cell type. Chemotherapy was started postoperatively and marked decrease of enhanced area was observed in the CT scan taken on the 9th of July. But the patient showed melena and was diagnosed as having rectal adenocarcinoma histologically by biopsied specimen. Case 2 was a 48-year-old male who had been suffering from left facial palsy and vertigo since August, 1981. Those symptoms gradually progressed. On the 4th of December, he was admitted to our clinic. On admission, neurologically, multiple cranial nerve involvement was noticed such as of the IIIrd, IVth, Vth and VIth nerves bilaterally, and the VIIth, IXth, Xth and XIth cranial nerves on the left side. Right hemihypalgesia, thermohypesthesia, dysmetria, dysdiadochokinesis, and cerebellar taxia were also seen. CT scan and left vertebral angiogram revealed a tumor in the pons and IVth ventricle. On December 8th, suboccipital craniectomy was performed, but almost all of the tumor seemed growing under the floor of the IVth ventricle.(ABSTRACT TRUNCATED AT 250 WORDS)

Decreased Na+-gradient-dependent D-glucose transport in brush-border membrane vesicles from rabbits with experimental Fanconi syndrome.

The effect of anhydro-4-epitetracycline on sodium gradient-dependent D-glucose transport of rabbit renal brush-border membrane vesicles was studied. The purity of isolated brush-border membrane vesicles as judged by enzyme activities was not different between normal control and anhydro-4-epitetracycline-administered rabbits. There was no difference in estimate of intravesicular volume, either. When NaCl was used for sodium gradient, the overshoot of D-glucose uptake into brush-border membrane vesicles isolated from anhydro-4-epitetracycline-treated rabbits was significantly smaller than that of normal control rabbits. In the cases of NaSCN or Na2SO4, the former was also smaller than the latter, but not significantly so. To avoid the possible effect of membrane potential on D-glucose uptake, the voltage-clamp method was applied. Even in the voltage-clamped condition, the overshoot of D-glucose uptake into vesicles from anhydro-4-epitetracycline-treated rabbits was decreased compared to that of normal rabbits. In vitro incubation of brush-border membrane vesicles with 20 mM anhydro-4-epitetracycline caused no alteration in sodium gradient-dependent D-glucose uptake. Our results demonstrate that there exists a disorder in sodium gradient-dependent D-glucose uptake of renal brush-border membrane in anhydro-4-epitetracycline-treated rabbits, and suggest that this disorder is one of the underlying mechanisms of experimental Fanconi syndrome.

Affinity of 167Tm-citrate for tumor and liver tissue.

Strong affinity of 167Tm-citrate for tumor tissue was reconfirmed by using Ehrlich tumor. Excellent tumor imaging was obtained with 167Tm-citrate because of its strong tumor affinity and because of the suitable physical characteristics of 167Tm. A large number of 167Tm had accumulated in the connective tissue which contained inflammatory tissue, quite large amounts were found in areas containing viable and necrotic tumor tissue, and small amounts were present in viable tumor tissue. 167Tm was not seen in necrotic tumor tissue. It was concluded that lysosomes did not play a major role in the tumor concentration of 167Tm, but played an important role in the liver concentration of this nuclide. In the case of hepatoma AH109A, it was presumed that lysosomes played a considerably important role in the tumor concentration of 167Tm, hepatoma AH109A possessing some residual features of the liver. 167Tm was bound to acid mucopolysaccharides and transposed by the acid mucopolysaccharides in the tumor tissues and liver. The acid mucopolysaccharides to which 167Tm were bound in tumor and liver, were heparan sulfate, chondroitin sulfate (or keratosulfate) and heparin (or keratosulfate).
