Arterio-venous fistulas constructed with formalin-fixed human umbilical grafts.

Since July 1976 we have implanted 18 umbilical vein grafts as haemodialysis shunts. Our experience shows that the umbilical vein is suitable for all shunt localisations. The formalin-fixed umbilical vein graft can be considered for vascular replacement. These can be removed in any clinic, prepared and stored under sterile conditions. The formalin conservation procedure is simple, inexpensive and possible everywhere without elaborate technical equipment.

Microcalorimetric investigations of the metabolism of isolated human epidermis.

Microcalorimetry was used for the evaluation of metabolic and enzyme activity in human epidermis. The endogenous metabolism with a heat production Q = -2 muW/mg wet weight could be stimulated to Q = -12 muW/mg by adding glucose to the buffer. To test the enzyme activity, hydrogenation of pyruvate to lactate by lactate dehydrogenase (LDH) was chosen as an essential step of anaerobic glycolysis. The LDH activity of epidermis slices amounted to 22 mU/mg epidermis. The microcalorimetric method is discussed as a test procedure for specimens of healthy and diseased skin and for cell cultures.

[Comparative immunological and morphological studies following transplantation of fresh and preserved allogeneic vein and artery segments in rat experiments].

Immunological and morphological investigations were carried out on immunogenetically different strain combinations of inbred rats following transplantation of fresh allogeneic and implantation of formalin preserved vein segments. In contrast to vital allogeneic transplants, which evoke an immunological response depending on immunogenetic difference between donor and recipient, there is neither humoral nor cellular immunological response after implantation of formalin fixed vein segments. Therefore, from an immunological standpoint, strongly allogeneic vein implants preserved by formalin can be used for replacement of arteries.

Orthotopic and heterotopic autotransplantation of the rat kidney.

End-to-end anastomosis of the kidney vessels and ureter can easily be done without suture. Artery and ureter are connected via an intraluminal silicone tube, anastomosis of the vein is performed with a cuff-technique. In case of orthotopic transplantation the left kidney is transplanted to the right side after right nephrectomy. Furthermore heterotopic transplantation was tried: kidney vessels were either connected with jugular or femoral vessels. In contrast to orthotopic transplantation higher thrombosis rate was observed, probably due to varying sizes of donor and recipient vessels. Our method of orthotopic autotransplantation is easier, less time consuming and has a lower complication rate than homologous kidney transplantation with suture techniques.

Genetics of susceptibility in the platyfish/swordtail tumor system to develop fibrosarcoma and rhabdomyosarcoma following treatment with N-methyl-N-nitrosourea (MNU).

About 7000 animals of 65 different genotypes of the xiphophorine fish were treated with the direct acting chemical carcinogen N-methyl-N-nitrosourea (MNU; 10(-3)M; four times for 1 hour in two week intervals), in order to find out whether the susceptibility for development of fibrosarcomas and rhabdomyosarcomas is directly related to the genotype. A genotype specific susceptibility was found, ranging from zero to about nine percent. The highest susceptibles were found in certain backcross hybrids involving P.variatus/X.helleri-hybrids and X.helleri, as the recurrent parent. These genotypes were further analysed. Both P.variatus and X.helleri, as werr as their F1 proved to be insusceptible; while from the three backcrosses, which were tested, namely the BC1, BC4 and BC15, both the BC1, and the BC4, were susceptible, but the BC15 was insusceptible. The results are interpreted on the basis of the assumption that the differential susceptibility is a function of the type of control of a tumor gene (Tu-Fi-Rh) endogenous to P.variatus and involved in development of fibrosarcomas and rhabdomyosarcomas. Accordingly, in P.variatus and in the F1 the Tu-Fi-Rh is controlled by repressing genes (R-genes) linked as well as non-linked to Tu-Fi-Rh; because simultaneous mutation of both R-genes following treatment with MNU is an extremely unlikely event, these genotypes have an extremely low susceptibility. By contrast, in the BC1 and the BC4 the non-linked R-genes become eliminated and only the linked R-gene remains for repression of Tu-Fi-Rh; this condition confers a high degree of susceptibility, because one single mutation may lead to impairment of the R-gene and to Tu-Fi-Rh-mediated formulation of fibrosarcomas and rhabdomysarcomas. In the BC15, furthermore, also the Tu-Fi-Rh has become eliminated, resulting in a loss of the susceptibility. The results suggest that in the xiphophophorine fish the susceptibility for responding to MNU-treatment with the development of fibrosarcomas and rhabdomysarcomas is related directly to the genotype.

[Formalin preserved allogenic veins for arterial replacement (author's transl)].

Allogenic saphena magna and umbilical vein grafts can be successfully used for vessel replacement and dialysis shunts. The chemical preservation with 4% buffered formalin is to be recommended as a preservation procedure, since the preparation of the vein grafts can take place aseptically and without the occurrence of immunologic rejections. The conservation procedure is simple, inexpensive and possible everywhere without elaborate technical equipment. The dissecting technique and preparation is demonstrated in detail.

[Implantation of formalin-preserved umbilical veins as dialysis shunts--a new method of allogenic blood vessel graft (author's transl)].

Allogenous venous umbilical implants can be used for successful vascular reconstructions and for the establishment of dialysis shunts. Presently 15 patients were treated with venous umbilical implants, in two of which a thrombosis of the implant occurred, one of them showed slight dilatation. In the case of dialysis shunts, implants serve exceptionally well. Especially the fixation in formalin can be especially recommended for preservation due to the fact that the umbilical veins can be obtained under non-sterile conditions, the immunological rejections are omitted. According to the present observations the mechanical properties are favourably influenced. At the time of the operation the umbilical veins do not possess an endothelium, nevertheless their functions are thereby affected. The results up to present time are favourable, nevertheless further investigations are necessitated.

[Value of formalin-fixed, allogeneic veins for vessel substitution in long-term tests. Experimental studies of the problem of immunological reactions on RtH-1-incompatible various rat-donor-recipient combinations].

Experiments on an immunogenetically standardized inbred rat strain combination were necessary to determine whether immunological reactions have an effect on the increase and transformation processes of allogeneic formalin-fixed venous implants. By using a RtH-1-incompatible allogeneic strain combination we found that immunological mechanisms-humoral or cellbound-are insignificant to such venous implants. The formalin-fixing procedure caused the antigenicity to lose its effectiveness. Thus, the submitted material shows that immunological reactions are not responsible for delayed alterations in such venous implants.

[False positive tumour diagnosis in the caecum(author's transl)].

Radiographic appearances simulating tumours were found in 25 patients; the causes of the mistaken diagnosis are critically analysed. The commonest mistake was caused by inflammatory process (eight patients), caecal contents (seven patients) and an enlarged ileo-caecal valve (five patients). Ways of avoiding such misinterpretation are described.author
