Protein kinase C-dependent regulation of sulfidopeptide leukotriene biosynthesis and leukotriene C4 synthase in neutrophilic HL-60 cells.

In response to calcium ionophore (A23187) stimulation, human granulocyte/macrophage colony-stimulating factor-primed, dimethylsulfoxide-differentiated HL-60 cells (which resemble mature granulocytes) synthesized leukotrienes (LTs) LTA4, LTB4, LTC4, and LTD4. The synthesis of the sulfidopeptide LTs, LTC4 and LTD4, was specifically inhibited in cells incubated in the presence of both A23187 and phorbol-12-myristate-13-acetate (PMA), an activator of protein kinase C (PKC). In contrast, neither the synthesis of LTB4, a product of the nonpeptide branch of the LT pathway, nor the formation of LTA4, the precursor for both branches of the LT biosynthetic pathway, was significantly affected by the presence of PMA during A23187 stimulation. The inhibition by PMA of LTC4 production in A23187-stimulated HL-60 cells was dose dependent, with an IC50 value of approximately 3.5 nM. The PKC inhibitor staurosporine completely reversed the inhibition by PMA of LTC4 production in A23187-stimulated cells, in a dose-dependent fashion, with an IC50 value of approximately 30 nM. Bisindolylmaleimide, another PKC inhibitor, was also able to prevent PMA-mediated inhibition of LTC4 formation, whereas inhibitors of protein kinase A, tyrosine kinases, or the respiratory-burst oxidase were not. Measurement of LTC4 synthase enzymatic activity in cells challenged with A23187 and PMA in the presence or absence of staurosporine demonstrated that the activity of the LTC4 synthase enzyme was inhibited in cells costimulated with A23187 and PMA and that inhibition could also be completely prevented by the presence of staurosporine. Because PMA is known to activate PKC, and staurosporine and bisindolylmaleimide are inhibitors of PKC, these results suggest that LTC4 synthase in HL-60 cells may be phosphoregulated.

The effect of hydroxyl radical on the antigenicity of native DNA.

Hydroxyl free-radical mediated in vitro modification of native calf thymus DNA showed single-strand breaks, decrease in melting temperature and structural alteration of purine and pyrimidine bases. Experimentally induced antibodies against modified DNA exhibited polyspecificity. Native DNA, RNA and synthetic polynucleotides in B-conformation were found to be an effective inhibitor of induced antibody-immunogen interaction. Naturally occurring human anti-DNA autoantibodies showed enhanced recognition of modified DNA as compared to native polymer. IgG isolated from anti-DNA antibody-positive SLE sera by Protein A-Sepharose 4B chromatography were affinity purified on nDNA-(polylysyl-Sepharose 4B) matrix. In competitive binding experiments, the affinity-isolated IgG showed almost equal recognition of both native DNA and hydroxyl radical modified DNA. The possible role of hydroxyl radicals in the etiopathogenesis of SLE has been discussed.

Purification to homogeneity and the N-terminal sequence of human leukotriene C4 synthase: a homodimeric glutathione S-transferase composed of 18-kDa subunits.

Human leukotriene C4 (LTC4) synthase was purified > 25,000-fold to homogeneity from the monocytic leukemia cell line THP-1. Beginning with taurocholate-solubilized microsomal membranes, LTC4 synthase was chromatographically resolved by (i) anion exchange, (ii) affinity chromatography (through a resin of biotinylated LTC2 immobilized on streptavidin-agarose), and then (iii) gel filtration. The final preparation contained only an 18-kDa polypeptide. The molecular mass of the pure polypeptide was consistent with an 18-kDa polypeptide from THP-1 cell membranes that was specifically photolabeled by an LTC4 photoaffinity probe, 125I-labeled azido-LTC4. On calibrated gel-filtration columns, purified LTC4 synthase activity eluted at a volume corresponding to 39.2 +/- 3.3 kDa (n = 12). The sequence of the N-terminal 35 amino acids was determined and found to be a unique sequence composed predominantly of hydrophobic amino acids and containing a consensus sequence for protein kinase C phosphorylation. We therefore conclude that human LTC4 synthase is a glutathione S-transferase composed of an 18-kDa polypeptide that is enzymatically active as a homodimer and may be phosphoregulated in vivo.

Photoaffinity labelling of human leukotriene C4 synthase in THP-1 cell membranes.

Human leukotriene C4 synthase specific activity in the human monocytic leukemia cell line THP-1 (0.302 +/- 0.062 nmol LTC4 formed.min-1 x mg-1) was 7.6-fold higher than in U937 cells (0.040 +/- 0.017 nmol LTC4 formed.min-1 x mg-1) and comparable to dimethylsulfoxide-differentiated U937 cells (0.399 +/- 0.084 nmol LTC4 formed.min-1 x mg-1). Using the photoaffinity probe, azido[125I]-LTC4, a single polypeptide with a molecular mass of 18 kDa was specifically labelled in THP-1 microsomal membranes. The rank order of potencies for competition of azido[125I]-LTC4 photolabelling of the 18 kDa protein by glutathione, leukotrienes and their analogs was found to be LTC2 > (azido[127I]-LTC4 approximately LTC4) > (LTD4 approximately LTE4) > (LTA4 approximately LTB4) > S-hexyl glutathione > glutathione, corresponded with the rank order of potencies for inhibition of LTC4 synthase activity but not inhibition of microsomal glutathione S-transferase activity. The 18 kDa protein specifically labelled by azido[125I]-LTC4 had high specificity for LTC4 and closely related leukotrienes and was separable from microsomal glutathione S-transferase. We conclude that azido[125I]-LTC4 specifically photolabels LTC4 synthase which is an 18 kDa polypeptide or contains an 18 kDa subunit.

Expression of endogenous and microinjected hsp 30 genes in early Xenopus laevis embryos.

In the present study, we have examined the regulation of expression of a newly isolated member of the hsp 30 gene family, hsp 30C. Using RT-PCR, we found that this gene was first heat-inducible at the tailbud stage of development. We also examined the expression of two microinjected modified hsp 30C gene constructs in Xenopus embryos. One of the constructs had 404 bp of hsp 30C 5'-flanking region, whereas the other had 3.6 kb. Both gene constructs had 1 kb of 3'-flanking region. RT-PCR assays were employed to detect the expression of these microinjected genes. The presence of extensive 5'- and 3'-flanking regions of the hsp 30C gene did not confer proper developmental regulation, since heat-inducible expression of both of the microinjected constructs was detectable at the midblastula stage. The premature expression of the microinjected hsp 30 gene was not a result of high plasmid copy number of the presence of plasmid DNA sequences. These results suggest that the microinjected genes contain all the cis-acting DNA sequences required for correct heat-inducible regulation but do not contain the elements required for the proper regulation of hsp 30 gene expression during development. It is possible that regulatory elements controlling the developmental expression of the hsp30 genes may reside upstream or downstream of the entire cluster.

Visceral leishmaniasis in Ethiopia. I. Cross-sectional leishmanin skin test in an endemic locality.

A cross-sectional leishmanin skin test was carried out on a sample population of 730 individuals. The overall prevalence of leishmanin positivity was 36.4%; 50.9% of males and 23.2% of females showed positive skin test reaction. The skin-test profile in the study area is typical of an endemic focus of visceral leishmaniasis. The increase in leishmanin positivity with age parallels the age-specific disease profile and indicates an outdoor exposure to infection.

Visceral leishmaniasis in Ethiopia. II. Annual leishmanin transformation in a population. Is positive leishmanin reaction a life-long phenomenon?

A one-year follow-up leishmanin skin test was undertaken in 605 subjects aged five years and above. The point prevalence of leishmanin positivity in the selected age group was 46.8%; 62.5% of the males and 32.1% of the females showed positive leishmanin reactions. The annual incidence of skin test transformation was 14.8%. In this study, positive to negative leishmanin conversion was noted in 9.3% of study subjects with initial positive leishmanin reaction.

Serial SPECT imaging in moyamoya using I-123 IMP. A method of noninvasive evaluation and follow-up.

Serial cerebral perfusion studies were performed in a patient with moyamoya disease, utilizing N-isopropyl iodoamphetamine (I-123 IMP). The SPECT findings correlated closely with those of a CT scan and magnetic resonance imaging (MRI). In fact, the perfusion defects seen on SPECT studies were larger than those seen on CT and MRI studies. The SPECT findings paralleled the patient's improving clinical course. Cerebral perfusion SPECT studies may be very helpful in the evaluation and follow-up of patients with moyamoya disease.

Dendritic cells infected in vitro with human T cell leukaemia/lymphoma virus type-1 (HTLV-1); enhanced lymphocytic proliferation and tropical spastic paraparesis.

Evidence supporting a role of the dendritic cell (DC) in stimulating autologous T cell activity in tropical spastic paraparesis (TSP) was sought by studies of cells taken from healthy volunteers and exposed to HTLV-1 in vitro. DC were co-cultured with an HTLV-1-producing cell line (MT-2) at 1:1 or 10:1 ratios. These DC stimulated high levels of proliferation in autologous T cells. This was similar to that seen in an autologous mixed leucocyte reaction (AMLR) using cells from TSP patients. The requirement for both DC and virus was confirmed, since neither DC co-cultured with uninfected MT-2 cells nor addition of infected MT-2 cells directly to T cells caused significant stimulation. DC exposed to the highest dose of HTLV-1 (1:1) for 24 h before addition of T cells caused strong stimulation that increased after 8 h but almost disappeared by 72 h. In situ hybridization showed that approximately 25% of DC became infected in cultured cells after preincubation for 24 h, and over 50% were infected with a 72-h preincubation. We suggest that infection of DC by HTLV-1 may be an initial step in altering the immune system in seronegative patients, and that persistent T cell stimulation in those with genetic susceptibility may underlie the production of neurological disease.

Genetic evidence for the role of isocytochrome c2 in photosynthetic growth of Rhodobacter sphaeroides Spd mutants.

In Rhodobacter sphaeroides, cytochrome c2 (cyt c2)-deficient mutants are photosynthetically incompetent (PS-). However, mutations which suppress the photosynthetic deficiency (spd mutations) of cyt c2 mutants increase the levels of a cyt c2 isoform, isocyt c2. To determine whether isocyt c2 was required for photosynthetic growth of Spd mutants, we used Tn5 mutagenesis to generate a PS- mutant (TP39) that lacks both cyt c2 and isocyt c2. DNA sequence analysis of wild-type DNA that restores isocyt c2 production and photosynthetic growth to TP39 indicates that it encodes the isocyt c2 structural gene, cycI. The Tn5 insertion in TP39 is approximately 1.5 kb upstream of cycI, and our results show that it is polar onto cycI. The cycI gene has been physically mapped to a region of chromosome I that is approximately 700 kb from the R. sphaeroides photosynthetic gene cluster. Construction of a defined cycI null mutant and complementation of several mutants with the cycI gene under the control of the cyt c2 promoter region indicate that an increase in the levels of isocyt c2 alone is necessary and sufficient for photosynthetic growth in the absence of cyt c2. The data are discussed in terms of the obligate role of isocyt c2 in cyt c2-independent photosynthesis of R. sphaeroides.

The distribution of medication along the spinal canal after chronic intrathecal administration.

Chronic intrathecal drug infusion for the treatment of neurological diseases, such as spasticity and chronic pain, has become an accepted method of therapy in recent years. Concurrent pharmacokinetic studies have shown that the cisternal cerebrospinal fluid (CSF) drug level is considerably lower than the lumbar CSF level during continuous infusion into the lumbar subarachnoid space. One factor that makes analysis of this decline in drug level difficult to quantify is that it is only feasible to sample CSF at the two extremes of the spinal subarachnoid space. Using a radionuclide technique, we have examined the distribution along the spinal canal of a hydrophilic compound, indium-111 diethylenetriamine pentaacetic acid, that was delivered over 72 hours into the lumbar subarachnoid space in five patients with implanted drug pumps. Over a 20-cm distance of the thoracic cord, radionuclide counts decreased gradually so that the indium-111 diethylenetriamine pentaacetic acid concentration surrounding the cord at the T2 vertebral level was 43% of that at the T12 level in four patients. Therefore, it appears that even with a hydrophilic compound, which minimizes spinal cord capillary losses, there is still a considerable reduction of CSF drug concentration along the spinal canal. The clinical implication of this gradual decline in drug level is that for intrathecal infusion of relatively hydrophilic compounds there may not be any advantage in placing the catheter tip at more rostral locations, such as at the midthoracic or cervical cord.

Risk factors and treatment of endometrial carcinoma.

A twelve-year retrospective study of 54 consecutive cases of endometrial carcinoma revealed that post-menopausal bleeding was the commonest symptom, and the major associated risk factors were obesity and hypertension. Panhysterectomy was the corner-stone of treatment while adjunctive therapy was based on certain prognostic factors and the operator's preference.

Cold-knife cervical conization.

Forty-six cervical conizations were performed over a ten-year period for diagnostic or therapeutic purposes. Twenty-nine women were under 45 years of age. The most common reason for conization was an abnormal Papanicolaou smear. The overall incidence of neoplasia was 80.4 per cent. The high complication rate of 28.4 per cent emphasizes the need for a less traumatic technique of excisional biopsy.

Efficacy of two formulations of the insect growth regulator, pyriproxyfen (Nylar or Sumilarv), against nuisance Chironomidae (Diptera) in man-made ponds.

A juvenoid insect growth regulator, pyriproxyfen (Nylar or Sumilarv), in a 10% emulsifiable concentrate (10 EC) and a granular (3% sand granules) formulation was evaluated for efficacy against natural populations of chironomid midges in man-made ponds in central Florida. Both formulations at 0.05 kg AI/ha induced complete suppression of adult midge emergence for 1 wk. At 0.2 kg AI/ha, emergence of adult midges completely ceased for 2 and 5 wk with the EC and the granules, respectively. The EC formulation was not effective beyond 2 wk, but the granules at both treatment rates reduced midge emergence by 81-100% for 9 wk posttreatment. The differences in suppression of adult midge emergence between the 2 formulations and times posttreatment were both highly significant (P < 0.01). No significant difference (P > 0.05) was found between the posttreatment percent inhibition of midge emergence caused by the lower and the higher rates of the granular formulation. Some reductions of midge larvae occurred in the ponds receiving the higher rate of the 2 formulations, but these reductions were statistically not significant.

Evaluation of a probe hybridisation serotyping method for group A rotavirus.

Serotype-specificity and sensitivity of oligonucleotide probes to serotype human rotaviruses was assessed. Probes could detect as little as 6.3 ng of homologous RNA and none reacted with as much as 100 ng of heterologous RNA. Northern-blot analysis revealed that probes reacted with one of genomic segments 7, 8 or 9 of corresponding serotypes.

Is adolescent pregnancy hazardous?

A study of the pregnancy outcome in 3320 teenagers revealed a low incidence of pre-eclampsia and anaemia and high rates of eclampsia, prematurity and low birthweight. The perinatal loss was 2.2 per cent and there was one maternal death. A comprehensive programme designed to improve antenatal surveillance is recommended for adolescents.
