[Effect of diazepam and nicotinamide on convulsive activity of various types].

Acute experiments on mice were made to investigate the anticonvulsant activity of diazepam and its combination with nicotinamide in experimental seizures induced by corazol, picrotoxin, bicucullin and thiosemicarbazide. It was shown that diazepam (0.2-1.6 mg/kg) produced a dose-dependent anticonvulsant effect in all models under study. Nicotinamide (250 mg/kg) substantially reduced the animals' lethality and seizure manifestations induced by picrotoxin and completely protected some of the animals from the bicucullin-induced seizures. Nicotinamide (250 mg/kg) potentiated the anticonvulsant effect of diazepam in all the experimental seizures under study. Micotinamide had a more pronounced effect in respect of the protection of the animals from tonic seizures as compared with clonic ones. The data suggest that the mechanisms of the anticonvulsant action of nicotinamide are mediated via the GABA-benzodiazepine receptor complex.

[Effect of nicotinamide on generalized convulsions in mice].

It was shown in acute experiments on mice that nicotinamide in doses of 250-500 mg/kg given intraperitoneally increased the latent period of corasole clonico-tonic convulsions without affecting their expressiveness and mortality. When given in a dose of 1000 mg/kg nicotinamide prevented clonico-tonic convulsions and mortality, and considerably raised the latent period of convulsions and the lifespan of the animals intoxicated with thiosemicarbazide and strychnine. The expressiveness of the nicotinamide effects depended on the time of its administration.

[Changes in the convulsant readiness of mice as affected by korazol].

It has been demonstrated in experiments on F1 (CBA X C57BL/6) hybrid mice and random-bred white mice that daily administration of corazol (30 mg/kg) led to the development of pharmacological kindling. The kindling was marked by a statistically significant reduction in the convulsant dose (CD50) of corazol, picrotoxin, bicuculline and thiosemicarbazide. No substantial differences were discovered in the degree of seizures induced by strychnine in the "kindling" and control groups. It is assumed that the mechanisms of the corazol kindling are effected via impairment of the GABA-benzodiazepine receptor complex.

[Interaction of excitatory foci and their complexes in the cerebral cortex].

Relations between excitation foci and their complexes in the cerebral cortex were studied in acute experiments on cats. Excitatory foci were produced by application of solutions of strychnine, penicillin and acetylcholine with prozerine in different concentrations. Two main types of interrelations were established: the determinant and the dominant ones. It was shown that a powerful excitation focus may enhance and synchronize the activity in weaker foci, integrate them in a single functional complex and determine the pattern of its activity (determinant relationships). It may also suppress the activity of previously created foci (dominant relations). The latter form of interrelations was observed either in the transitional stage during formation of the complex or it had an independent meaning. Such types of functional interrelations were observed both between separate foci and their complexes.

[Effect of pyridoxal-5-phosphate on epileptic activity in the cerebral cortex].

It has been shown in acute experiments on cats that pyridoxal-5-phosphate (PALPh) injected intravenously in doses of 5-20 mg/kg suppresses the epileptic activity in solitary foci caused by penicillin and strychnine local application as well as in the complex of loci created in different zones of the brain cortex. Under the influence of PALPh the complex was destabilized and suppressed, and the epileptic activity first disappeared in the dependent foci and then in a determinant one. It is concluded that PALPh has antiepileptic activity. This effect seems to be more pronounced in the penicillin-induced epileptic foci.

[The role of the substantia nigra in the mechanisms of the cessation of epileptic activity].

In the review of literature and author's own data the participation of pars compacta and pars reticulata of the substantia nigra in mechanisms of epileptic syndrome development and cessation of epileptic activity was shown. The results are given which testified for dependence of substantia nigra-derived effects upon form and intensity of epileptic activity as well as upon neuromediator and peptidergic systems of the brain involvement. The conclusion was made concerning the significance of substantia nigra as a part of antiepileptic system of the brain in the processes of restriction and suppression of epileptiform manifestations.
