[Characteristics of hemagglutination reaction in methanotrophic bacteria].

The reaction of hemagglutination with trypsin-treated rabbit erythrocytes was used to reveal lectins on the cell surface of methanotrophic bacteria and in their culture liquids. By this method, no lectins were detected on the cell surface of Methylococcus capsulatus IMV B-3001 and Methylomonas rubra IMV B-3075 or in the culture liquid of any of the species studied. With intact cells of Methylocystis parvus IMV B-3491, the positive hemagglutination reaction observed was nonspecific and most probably occurred due to the high cell surface hydrophobicity characteristic of this species.

[New strains of soil bacilli which mineralize organic phosphorus compounds].

Microorganisms, which mineralize organophosphates (216 isolates) have been isolated for the chernozem soils of different regions of Ukraine. Sixteen most active strains referred to Bacillus genus have been identified and studied. These microorganisms grow (with different intensity) on the mineral medium with glucose and glycerophosphate or phytin, accumulating phosphate-ions in the medium. The most active strains mineralized glycerophosphate, accumulating about 280 micrograms/ml of PO4(3-) and phytin accumulating up to 130 micrograms/ml of PO4(3-) during 3 days. New strains of bacilli have been chosen for creation of combined bacterial preparations of the complex effect.

[Physiological activity of mixed cultures of Methylcoccus capsulatus UKM B-3030 with Bacillus megaterium UKM B-5723T and Bacillus subtilis VKPM B-1489 on solid surface colonization].

Physiological activity of monoculture of Methylococcus capsulatus UCM B-3030 and its mixed cultures with two bacilli species distinguished by proteolytical properties--Bacillus megaterium UCM B-5723T and Bacillus subtilis BK[symbol: see text]M B-4189 in terms of long-duration cultivation is investigated. It is shown, that the most active methane consumption by methanotrophic bacteria and their mixed cultures with bacilli under solid surface colonization occurred on the second-eighth day of cultivation. The obtained results permit one to evaluate intensity of CH4 oxidation process by typical microflora of some econiches (depleted coal mines, in particular).

[Features of solid materials' colonization by pure and mixed cultures of methanotrophs].

The process of colonization of hydrophilic (glass) and hydrophobic (polysterene) carriers by pure cultures of methanotrophs Methylocystis parvus UCM B-3490T, Methylococcus capsulatus UCM B-3030, as well as by their cultures mixed with Bacillus megaterium UCM B 5723T and Pseudomonas putida VKPM B-4188 under the conditions efficient for methanotrophic bacteria. M. parvus demonstrated the highest intensity of this process on the above carriers owing to high hydrophobic cell surface. Both methanotrophs colonized the glass surface more quickly with formation of microcolonies on carriers after 6 days of incubation in pure and mixed cultures with B. megaterium. The number of bacilli on these carriers quickly decreased. In the mixed cultures with P. putida the glass and polysterene colonization intensity decreased, while the amount of pseudomonas on carriers increased.

[Biological properties of the phosphate-mobilizing Bacillus subtilis strain IMV V-7023].

Biological characteristics of a new phosphate-mobilizing bacillus strain are reported. Species-level identification of the strain was performed according to morphological, cultural, and biochemical characteristics and the sequence of the 16S rRNA gene. The strain was identified as Bacillus subtilis IMV V-7023 and displayed a very high ability to mobilize phosphorus from its sparingly soluble inorganic and organic compounds and the capability of synthesizing biologically active substances; in addition, the strain essentially suppressed the growth of phytopathogenic bacteria, micromycetes, and agents causing various diseases of vegetable, cereal, leguminous, and other plants. The strain Bacillus subtilis IMV V-7023 is promising for developing bacterial preparations for crop production.

[Soil microflora over the closed Donbas mines].

Composition of soil air and microflora in different soil horizons (to 30 cm depth) over some closed Donbas coal mines has been studied. The intensive methane leakages have been found in some soil zones over the closed coal mines. This results in formation of explosive and inflammable gas mixtures. Methane content in soil gas mixtures could vary from trace amounts to 34 %. The essential decrease of oxygen (to 3-4%) and increasing (up to 5 %) of CO2 concentrations that evidences for active microbiological processes in such soils is marked in the latter case. Methanotrophic bacteria involving methane into biogenic cycle are widespread (up to 106 cells per 1 g) in methane-containing soils over the closed coal mines. Microorganisms of different physiological groups in the community with methanotrophic bacteria were found in the given technogenic econiche.

[Effect of cultivation conditions of Bacillus subtilis IMV B-7023 on surface properties of cells].

It has been established that the cells of Bacillus subtilis IMV B-7023 grown on the medium where peptone or calcium glycerophosphate served as the only source of carbon and energy have similar acid type of the surface and are characterized by a high negative charge even at low values of pH of the dispersion medium. This may be determined by the presence of phosphate groups, teichoic acids being their carriers in Gram-positive bacteria. Glucose addition to peptone or glycerophosphate leads to the change of the bacteria surface type, i.e., at low values of pH the cells have total positive charge, that allows to suppose availability of protein nature components in the surface layers of the cell wall. Bacteria grown in these conditions are also characterized by the higher degree of adhesion to inert carrier (glass).

[Characteristics of spore-forming bacteria of the genus Bacillus that break down caprolactam].

Five strains of sporeforming bacteria were isolated from sewage of capron industry and their morphological, cultural, as well as physiological and biochemical properties were investigated. Four strains were identified as Bacillus subtilis and one as Bacillus pumilus. The cultures were able to grow in mineral medium with caprolactam as a source of carbon and nitrogen. The influence of growth conditions on the rate of caprolactam decomposition in synthetic medium was studied.

[Microbial contamination in a water-treatment system].

Microorganisms destructors of organic compounds contained in the Kremenchug reservoir and water treatment system of heat power station have been distinguished and identified. This system does not completely remove microflora at all stages of technological process that results in infestation of ion-exchange resins of the 1st stage filters for water desalting with microorganisms. The reasons have been established of the 1st stage desalting filters infestation by the reservoir water microflora. It is necessary to form a complex approach to solution of the problem of preventing the accident situations: microbe infestation of ion-exchange resins of water-desalting filters in the system of chemical water treatment of heat power stations.
