[Polymerase and immunologic activity of reverse transcriptase of the HIV-1 virus, isolated from Escherichia coli].

The recombinant reverse transcriptase of HIV-1 virus has been isolated from Escherichia coli cells transformed by the plasmid pRT40 DNA. The 103 Kd protein produced by these cells is shown to be processed to proteins with lower molecular masses by the reverse transcriptases own protease activity as well as Escherichia coli proteases. The resulting 103-66 Kd proteins possess the polymerase activity while 51 Kd and smaller proteins are lacking the activity. The 66 and 51 Kd reverse transcriptase fragments demonstrate the positive immunological reaction with the human blood serum from the people possessing antibodies to HIV-1 virus. The recombinant reverse transcriptase of HIV-1 produced by Escherichia coli cells is shown to be useful in AIDS diagnosis in humans.

Recombinant reverse transcriptase of Rous sarcoma virus: characterization of DNA polymerase and RNAase H activities.

Enzyme preparations of Rous sarcoma virus (RSV) reverse transcriptase have been isolated from a culture of E. coli HB101(pMF14). The enzyme has been purified to homogeneity and been shown to consist of two subunits, of molecular mass 97.4 and 61.3 kDa, respectively. The optimum conditions for the DNA polymerase and RNAase H activities, fidelity of DNA synthesis on a homogeneous RNA template, and the inhibitory effect of azidothymidine triphosphate have been determined. Data on the use of RSV recombinant reverse transcriptase for cDNA synthesis are given.

[Recombinant RNA-dependent DNA-polymerase from Rous sarcoma virus. Isolation and properties].

Preparations of recombinant reverse transcriptase RSV were isolated from E. coli HB101/pMF14 cell cultures. The enzyme purified to homogeneity was shown to be made up of two subunits with molecular masses of 97 +/- 4 and 61 +/- 3 kDa. A comparison of enzymatic properties of recombinant transcriptase to those of the enzyme isolated from the RSV (Rauss sarcoma) virus demonstrated that in the preparations under study the recombinant reverse transcriptase exists in a subunit form, alpha beta, and may acquire a relatively stable configuration, alpha 2.

[Hyperbaric oxygenation in the combined therapy of glomerulonephritis].

As many as 51 patients afflicted with glomerulonephritis were examined. Of these, 26 received multimodality treatment including hyperbaric oxygenation (HBO) sessions. The efficacy of the multimodality treatment was assessed from the totality of the clinico-biochemical parameters, measurements of the activity of lipid peroxidation, appraisal of the condition of the antioxidant body systems, the activity of A2 phospholipase and the lipid phase of erythrocytic membranes. The studies performed have demonstrated the clinical efficacy of HBO as well as positive changes in the structure of the lipid phase of cell membranes. To prevent unfavourable effects of HBO, it is suggested that the level of alpha-tocopherol in cell membranes should be controlled and the intake of the drugs potentiating the hyperoxic effect should be restricted.

[Transducing properties of lambda phagemids].

Transduction of genetic markers conferring drug-resistance with lambda phagemids has been studied in terms of the influence of various genetic conditions on the level of transduction. For this purpose, we have constructed and analysed a new series of phagemids - lambda gt::pBR322 and lambda gt::pUC19, in addition to earlier published phagemids. Experimental data indicate that the transduction frequency depends on the function of cI repressor and bacterial recA system, the plasmid orientation in the phagemid DNA, the selective marker used in an experiment and the concentration of the antibiotic in the medium.

[Relationship between immunogenic markers with metabolic processes in chronic pyelonephritis].

Such HLA phenotypes as A3B35 and A9B35 occur most frequently in patients with chronic pyelonephritis (CP). In these phenotypes there are most pronounced changes in the lipid phase of red cell membranes and renal tissue. The above changes of blood cell cytomembranes and renal tissue can be used as diagnostic criteria of inflammation activity in pyelonephritis while the HLA antigens are helpful for detection of CP risk groups.
