[Biological activity of different forms of bacteriophage lambda DNA].

Hershey circles and linear tandem aggregated forms of DNA have been obtained in vitro and treated with polynucleotide ligase to form phosphodiester bond. Using zone centrifugation in glycerol gradient covalently closed circles and linear dimers have been purified and their biological activity investigated. It was found that closed circular molecules lost most, if not all, of their activity in CaCl2-dependent system. In order to investigate the biological activity of tandem dimer molecules, hybrid dimers consisting of DNA's from lambda C1857 and lambda 1434 have been obtained. In plaque assay with the appropriate non-permissive strains of E. coli the efficiency of infectivity of hybrid dimers was measured. Biological activity of dimer molecules sealed with ligase was about 5% of the activity of linear monomers. Ig has been suggested that tandem dimers of lambda DNA joined by phosphodiester bond are able to penetrate into the CaCl2-treated host cells and both components of dimers are active during subsequent multiplication.

[Recombinant murine cell lines transformed by various vectors based on bovine papillomavirus type 1 and expressing human tissue plasminogen activator].

We have constructed a number of vectors which include transcriptional unit of human tPA cDNA and 100% BPV-1 DNA or 100% Lx DNA (mutant BPV variant with tandem duplication of LCR-E6-E7 region). Additional HSV-1 Tk-promoter was inserted in the flanks of viral DNAa in a set of constructions. A number of recombinant cell lines have been established by means of transformation using the constructed vectors. The increased focus formation activity and the improved vector properties were demonstrated for vector construction which included Lx DNA with additional Tk promoter for activation of early viral transcription. The possibilities of BPV-based vectors design are discussed.

[The existence of the ribonucleoprotein branching enzyme in rabbit skeletal muscle and ribozyme corresponding to it].

Amylose isomerase (AI) preparations were isolated from rabbit muscles after Petrova et al., as well as by the additional fractionation steps. Their homogeneity, enzymatic activity and RNA, isolated from those preparations, were characterized. AI preparations, as described by Petrova et al., proved to be heterogeneous in respect to the protein and RNA; by using additional fractionation methods RNA and protein have been separated from each other, which proves that a homogeneous stable ribonucleoprotein complex, exerting AI activity, does not exist. It was shown by three independent methods that AI preparations isolated after Petrova do not display branching, but have amylolytic activity. RNA, isolated along with the AI preparations, proved to be mainly total tRNA degraded to different degrees. No RNA corresponding to the previously sequenced 2.5S RNA could be detected in these preparations. RNA preparations do not manifest neither branching, nor amylolytic activity. Our data prove that there is no ribozyme, whose existence has been suggested previously.

[Expression of human tissue plasminogen activator by recombinant cell lines from various animals].

A number of recombinant cell lines which produce human tissue plasminogen activator (tPA) was obtained using different hosts--mouse, rat, hamster, simian and human cell lines. All types of recombinant lines secreted active r-tPA into conditioned medium. A slight difference between molecular weights of secreted variants of r-tPA was mediated by the different mechanisms of protein modification. Treatment of some recombinant cell lines with different substances resulted in increased levels of r-tPA production.

[Cloning and regulation of gene expression of EcoRV restriction- modification system].

A number of recombinant plasmids, containing EcoRV restriction-modification genes have been constructed. Individual genes of this system were introduced into plasmids of various incompatibility groups. Promoter regions of genes encoding methylase and restrictase have been cloned and studied. With the use of specialized vector pVE8 it was shown that the efficiency of the endonuclease gene promoter is comparable with early lambda phage promoters and produced about 70% of PL efficiency. The efficiency of the methylase gene promoter region was twice less than the efficiency of the restriction endonuclease gene promoter. Plasmid with restriction endonuclease gene promoter located downstream in relation to the additional regulatable phage lambda promoter PL has been obtained. It enabled us to construct strains 30-40 fold overproducing this enzyme under conditions of inactivation of the temperature sensitive phage repressor c1857. This construction directs the production of a high level (10%) of the total cellular soluble proteins) of the EcoRV restriction enzyme. The factors that influenced the level of enzyme synthesis under induction are discussed.

[Primary structure of RNA 3 of barley stripe mosaic virus and its variability].

The complete nucleotide sequence was determined for three variants of the third genomic component of BSMV strain Argentina mild. The common variant, RNA 3 (2797 nucleotide), contains two open reading frames (ORFs) coding for two proteins with Mr of 74,229 (putative BSMV RNA polymerase) and Mr of 16,994. The second ORF is expressed from a subgenomic RNA. The extended variant RNA 3 differs from the common one only by the presence of a direct tandem repeat 351-363 nucleotides in length (with some variability) encompassing part of the leader sequence and the beginning of the first ORF. The resulting protein has a Mr of about 86,000. The defective variant, RNA 4, carries a deletion of 185 nucleotides in the 3'-end proximal part of the first ORF, which shortens the product to a Mr of 60,344.

[Intragenomic polymorphism of the primary structure of 5S rRNA gene variants of the loach (Misgurnus fossilis L.). Determination of the transcriptional activity].

The primary structure of 12 cloned repeats of loach oocyte 5S rRNA genes was determined. The heterogeneity of nucleotide sequences was revealed in the coding regions and spacer of the genes. The results of the study on in vivo transcriptional activity of the cloned 5S rRNA gene variants are consistent with the localisation of site specific base substitutions in the coding part affecting the transcription. We have compared the nucleotide sequences of loach 5S rRNA gene variants and of Xenopus laevis, X. borealis and Bombyx mori 5S genes which can be actively transcribed in X. laevis oocyte nuclei. As a result we could propose a consensus nucleotide sequence in the internal control region (from 45-th up to 100-th nucleotide) of the eukaryotic 5S rRNA gene. This sequence comprises a RNA-polymerase III promotor and stretches interacting with transcriptional factors. We have considered the base substitutions in the nucleotide sequences of 5S gene variants exerted on the experimental model of loach 5S rRNA secondary structure. All base substitutions in actively transcribed genes do not influence the general double-stranded structures of the transcripts. However in 5S RNA transcripts from genes with low transcriptional activity base substitutions affecting the box c RNA-polymerase III promoter destroy hairpin II interacting with ribosomal proteins. We have concluded that two factors can restrict the divergency of 5S rRNA genes: (1) conservation of the nucleotide sequence in the gene internal control region, and (2) conservation of the general double stranded structures in 5S rRNA transcripts.

[Secondary structure of pre-rRNA 41S spacers and its possible role in determining the processing sites].

The mechanism of pre-rRNA processing is still unknown. In this paper we discuss a possible role of the secondary structure of the internal transcribed spacer region (ITS-1 and ITS-2) in processing of 41S pre-rRNA. Potential possibilities for double strand structure formation in ITS-1 and ITS-2 and its rearrangement in the course of processing of 41S pre-rRNA with involvement of U3 RNA were analyzed for a number of species. We have concluded that this rearrangement is the necessary stage in 41S pre-rRNA processing when every functionally designated area (for mature 18S, 28S and 5,8S rRNAs, for ITS-1, ITS-2 and 21S pre-rRNA) has to be organized into an individual domain. In these strongly structured domains 5'- and 3'-ends are spatially brought together whereas processing sites are localized between these compactised areas.

[Expression of human growth hormone in cultured mouse fibroblasts].

The new system for the transfer and expression of foreign genes based on retroviral vectors pPS-neo, conferring neomycin resistance was constructed. The BALB/c mouse cell lines producing highly active human growth hormone (more than 7 micrograms/ml into culture medium) were constructed using these vectors. An antibody column was used to purify the growth hormone from cell culture medium. Possibilities of producers to be applied for gene therapy are discussed.

[Study of the structure of phage lambda operator OR1 using two-dimensional 1H-NMR-spectroscopy].

NOESY spectroscopy at 500 HMz was employed to assign resonances of nonexchangeable protons in the 1H NMR spectrum of the synthesized 17 base pair duplex of deoxyribonucleotides comprising the OR1 operator, one of the specific binding sites for the bacteriophage lambda cro repressor. A pure absorption spectrum that was obtained by the phase sensitive detection technique allowed to perform a resonance assignment of base and deoxyribose protons, excepting H-5'- and H-5"-protons, on the basis of a single NOESY experiment, mixing time 200 ms. The sequential assignment strategy for 2D NMR spectra of oligonucleotides was used for this purpose. The data obtained and the previous results on OR3 are discussed in the view of the conformation of operators in solution. It is shown that both duplexes exist in the DNA B-form with its intrinsic anti conformation of the nucleotides. Conformation of DNA segments with identical primary structures are similar, and local deviations from the classical B-form are determined by a nucleotide sequence.

[A system of EcoRV restriction-modification: genes, enzymes and synthetic substrates].

The genes, encoding the restriction endonuclease and modification methylase EcoRV have been cloned from the natural plasmid pLG13 into pBR32 derivative vector pIL233. A resultant clone, expressing both enzyme activities, was used as a source of DNA for sequencing these genes by a procedure, that employed construction of deletion derivatives used to locate borders (by means of a functional test) and to sequence ca. 300 bp near the deletion breakpoint. From the sequence data, we infer that the endonuclease, a 29 KDa protein, and the methylase, a 36 KDa protein, are transcribed from a 310 bp intergenic region in opposite directions. There is no apparent homology between the enzymes and genes of the EcoRI and the EcoRV systems. A synthetic decamer, containing the EcoRV endonuclease recognition sequence and a phosphoamide bond at the cleavage point, is not cleaved by the highly purified endonuclease; the unmodified synthetic decamer is cleaved at the same conditions, only that the cleavage occurs to produce a blunt end--GAT/ATC, and not in a place previously reported (GATAT/C).

[Primary structure of cDNA for bovine beta-casein].

In this work the complete sequence of 1097 nucleotide bovine beta-casein cDNA has been determined. Sequencing of end labeled fragments was performed using the method of Maxam and Gilbert. Bovine beta-casein cDNA consist of a 672 nucleotide coding region, flanked by 60 nucleotide 5' and 358 nucleotide 3'-noncoding region. The restriction map of beta-casein cDNA was constructed. The computer analysis was done and the comparisons of the complete sequence of bovine beta-casein cDNA with other sequences, coding caseins in bovine, rat and guinea-pig was performed. It was determined, that cloned cDNA is related to genetic variant A1.

[Genetic engineering of peptide hormones. II. Possible polymorphism of bovine preprolactin. Molecular cloning data].

The primary structure of an insert from a clone isolated from the bovine pituitary cDNA library by hybridization with prolactin-specific probe has been determined. It was found that the rearrangement of cDNA took place in the process of cloning. The rearrangement includes the inversion of 5'-terminal and the deletion of the central part of cDNA. However from the structure of the insert we were able to deduce the sequences of 5'- and 3'-terminal regions of bovine preprolactin mRNA (257 and 551 bases long). The comparison of these sequences with those published earlier revealed several differences in the primary structure. The most essential of them is the additional triplet coding for alanine in position of -22 of the signal peptide. The heterogeneity of bovine preprolactin mRNA in the region coding for the signal peptide is considered to be a consequence of alternative splicing as it was shown for rat preprolactin mRNA.
